Metabolomics Study of Joint Synovial Fluid in Beagle Dogs

摘要: 目 的 基于代谢图谱作为量化关节腔滑液内代谢物的工具,发现比格犬关节发育过程中的代谢规律。方法 收集幼年和成年比格犬关节滑液,运用液质联用( LC-MS) 非靶向代谢组学手段,分析不同年龄段比格犬关节腔滑液代谢组学特征及其与发育的相关性。 结果 与同性别成年比格犬相比,幼年雄性比格犬关节滑液中参与蛋白质、氨基酸、矿物质代谢和 ABC 转运的代谢物含量较高;幼年雌性比格犬中与氨基酸、碳水化合物代谢和 ABC转运相关的代谢物含量较高。 结论 氨基酸、蛋白质、矿物质及碳水化合物的代谢能力与犬骨关节发育及关节健康密切相关。

Abstract: Objective To analyze and compare metabolomics characteristics of joint synovial fluid between juvenile and adult beagle dogs. Method The joint synovial fluids were collected from 18 juvenile and 16 adult beagle dogs. Samples were subjected to liquid chromatography and mass spectrometry ( LC-MS) -based nontargeted metabolomics analyses. Result Compared with that of adult dogs, male juvenile animals showed higher levels of metabolites related to amino acid biosynthesis, protein digestion and absorption, mineral absorption, and ABC transporter. In female animals, the contents of metabolites related to amino acid, carbohydrate metabolism and ABC transport were higher in juvenile than adult dogs. Conclusion The bone joint development and health are closely related to metabolic events of amino acids, proteins, minerals and carbohydrates.
